چکیده

Abstract TFE3 -translocation renal cell carcinoma ( -tRCC) is a rare and heterogeneous subtype of kidney cancer with no standard treatment for advanced disease. We describe comprehensive molecular characteristics 63 untreated primary -tRCCs based on whole-exome RNA sequencing. -tRCC highly heterogeneous, both clinicopathologically genotypically. ASPSCR1-TFE3 fusion several somatic copy number alterations, including the loss 22q, are associated aggressive features poor outcomes. Apart from tumors MED15-TFE3 fusion, most exhibit low PD-L1 expression T-cell infiltration. Unsupervised transcriptomic analysis reveals five clusters distinct angiogenesis, stroma, proliferation KRAS down signatures, which show association patterns prognosis. In line nature, high angiogenesis/stroma/proliferation cluster exclusively consists fusion. Here, we genomic provide insights into precision medicine this

Newly Designed Break-Apart and ASPL-TFE3 Dual-Fusion FISH Assay Are Useful in Diagnosing Xp11.2 Translocation Renal Cell Carcinoma and ASPL-TFE3 Renal Cell Carcinoma

The diagnosis of Xp11.2 translocation renal cell carcinoma (tRCC), which relies on morphology and immunohistochemistry (IHC), is often either missed in the diagnosis or misdiagnosed. To improve the accuracy of diagnosis of Xp11.2 tRCC and ASPL-TFE3 renal cell carcinoma (RCC), we investigated newly designed fluorescence in situ hybridization (FISH) probes (diagnostic accuracy study).Based on the...

Identification of molecular tumor markers in renal cell carcinomas with TFE3 protein expression by RNA sequencing.

TFE3 translocation renal cell carcinoma (tRCC) is defined by chromosomal translocations involving the TFE3 transcription factor at chromosome Xp11.2. Genetically proven TFE3 tRCCs have a broad histologic spectrum with overlapping features to other renal tumor subtypes. In this study, we aimed for characterizing RCC with TFE3 protein expression. Using next-generation whole transcriptome sequenci...

Next-generation sequencing of translocation renal cell carcinoma reveals novel RNA splicing partners and frequent mutations of chromatin-remodeling genes.

PURPOSE MITF/TFE translocation renal cell carcinoma (TRCC) is a rare subtype of kidney cancer. Its incidence and the genome-wide characterization of its genetic origin have not been fully elucidated. EXPERIMENTAL DESIGN We performed RNA and exome sequencing on an exploratory set of TRCC (n = 7), and validated our findings using The Cancer Genome Atlas (TCGA) clear-cell RCC (ccRCC) dataset (n ...
